Who developed retatrutide, and the sequence that got it to phase 3

Published 24 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
Retatrutide was discovered and developed by Eli Lilly, which also developed the dual GIP and GLP-1 agonist that preceded it. The public record runs from the discovery paper in Cell Metabolism and the phase 1b trial in The Lancet in 2022, through the two phase 2 trials in 2023 and the liver-fat substudy in 2024, into a phase 3 programme named TRIUMPH.

Discovery and early clinical

Coskun and colleagues at the sponsor published the discovery and preclinical characterisation in Cell Metabolism in 2022 under the development code LY3437943. The same year, Urva and colleagues published the phase 1b multiple-ascending-dose trial in people with type 2 diabetes in The Lancet.

Those two papers established the molecule's design, its receptor profile and its pharmacokinetics in people.

Phase 2

In 2023 Jastreboff and colleagues published the 48-week phase 2 obesity trial in the New England Journal of Medicine, and Rosenstock and colleagues the 36-week phase 2 type 2 diabetes trial in The Lancet. In 2024 Sanyal and colleagues published the MRI liver-fat substudy of the obesity trial in Nature Medicine.

By that point the compound had acquired its nonproprietary name and the publications use it.

Phase 3 and status

The phase 3 programme, TRIUMPH, comprises multiple large trials. Results are released by the sponsor as trials complete and will be published in due course; this site reports them when they are citable. No regulatory approval has been granted.

Retatrutide is supplied by REVIVE LAB UAE as an investigational research material for laboratory use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not approved for any use anywhere, and no dosing, administration or protocol guidance is provided.
