What GHRH looks like near a membrane, which residues matter, and what that says about the analogue

Like the other peptides of the glucagon-secretin family, growth hormone-releasing hormone has little structure in dilute water and adopts an alpha helix when it meets a membrane or a membrane-mimetic solvent, with hydrophobic residues on one face and polar ones on the other. Alanine-scanning and truncation studies through the 1980s and 1990s showed that the amino-terminal residues — tyrosine 1, alanine 2, aspartate 3 — are required for receptor activation, that the helical region from about residue 6 to 29 carries binding, and that residues 30 to 44 add affinity and stability. Tesamorelin leaves all of this intact and adds the hexenoyl group outside the activating residues.

The helix

Circular dichroism in water shows a mostly disordered peptide; in trifluoroethanol or at a membrane surface the helical content rises sharply. The helix is amphipathic, so it is thought to lie on the membrane surface and then present its amino terminus to the receptor core. This membrane-catalysed binding model is shared across the family and is one reason the carboxyl-terminal residues, which extend the helix, raise affinity.

The helix also explains why some analogues with helix-stabilising substitutions bind more tightly.

Which residues matter

Removing tyrosine 1 or altering alanine 2 and aspartate 3 abolishes activation while sometimes sparing binding, which is the basis of GHRH antagonists. Substitutions in the helix that preserve its amphipathic character are tolerated; those that break it are not. The carboxyl terminus can be removed to residue 29 with full activity retained, and the natural 40-residue form is as active as the 44.

The hexenoyl group sits on the alpha-amino group of tyrosine 1, blocking the enzyme without changing the residue the receptor reads.

Why it is here

Because the structural biology explains both the design of the analogue and the pharmacology of the class. It describes chemistry and no use of the pack.

Tesamorelin is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
