The releasing hormone as a sleep-promoting signal, as the 1990s work showed, and the boundary of what it establishes

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
Growth hormone-releasing hormone acts in the hypothalamus and basal forebrain as a sleep-promoting signal as well as a pituitary releasing factor. In animals, injection of GHRH increases non-REM sleep and its antagonists reduce it; in humans, Steiger and colleagues reported in 1992 that pulsatile intravenous GHRH increased slow-wave sleep in young men while somatostatin reduced it. The coupling explains why the largest growth hormone pulse coincides with the first slow-wave period: one hypothalamic signal drives both. The finding is part of the physiology of the hormone and of the ageing of sleep, and the Tesamorelin sleep pages on this site cite the related literature for the analogue.

The animal and human work

Rodent studies through the late 1980s and 1990s, many from the Obál and Krueger laboratories, showed that GHRH given centrally increased slow-wave sleep and that blocking it with antibodies or antagonists reduced sleep, independently of growth hormone itself. The 1992 human study used repeated intravenous pulses during the night and polysomnography, finding more slow-wave sleep and more growth hormone, with somatostatin producing the opposite. Later work in older adults found smaller effects, consistent with the age-related decline of both.

GHRH-deficient and receptor-deficient mice sleep less deeply, which supports the physiological role.

What it establishes and what it does not

That GHRH is one of the signals that regulate sleep depth and that the growth hormone pulse and slow-wave sleep share a driver. It does not establish that any GHRH analogue given for any purpose improves sleep in any population; the tesamorelin trials did not measure sleep as a primary endpoint, and the one cognition trial recorded sleep as a secondary measure. The physiology is firm; the application is not.

The older Tesamorelin sleep pages on this site predate this register and are read in that light.

Why it is here

Because the sleep physiology of the hormone is established and frequently invoked, and a reader should know its basis and its boundary. It describes physiology and no use of the pack.

Tesamorelin is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
