Why the hypothalamic releasing hormone was isolated from the pancreas, and who did it

The existence of a hypothalamic factor releasing growth hormone was inferred from the 1960s, but the hypothalamus contains too little to purify. In 1982 a patient with acromegaly caused by a pancreatic tumour secreting the factor gave the groups of Roger Guillemin at the Salk Institute and Wylie Vale with Jean Rivier the material they needed; Guillemin and colleagues published a 44-residue peptide in Science and Rivier and colleagues a 40-residue form in Nature the same year. The sequences matched the hypothalamic hormone when it was later obtained, and synthetic peptide released growth hormone in humans. Tesamorelin is the 44-residue form with one modification.

The search

Guillemin and Schally had shared a Nobel Prize in 1977 for isolating the thyrotropin- and gonadotropin-releasing hormones and somatostatin from hundreds of thousands of animal hypothalami. The growth hormone-releasing factor resisted the same approach because it is scarce and labile. Ectopic production by a tumour, in milligram amounts, solved the problem that brute-force extraction could not.

Two patients, one in the United States and one in France, supplied the tumours used by the two groups.

What was found

A 44-residue amidated peptide and a 40-residue carboxyl-terminally truncated form, both active. The first 29 residues were shown soon after to carry full activity, which gave the sermorelin fragment. The hormone belonged to the glucagon-secretin family by sequence. Synthetic hormone given to healthy volunteers released growth hormone promptly, confirming the identity.

The receptor was cloned a decade later.

Why it is here

Because the molecule in the pack is this hormone, and its discovery through disease is a part of its history worth stating. It describes history and no use of the pack.

Tesamorelin is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
