What the approved label lists, how often, and which reactions trace to the released growth hormone

The label groups the adverse reactions seen in the pooled phase 3 trials: injection-site reactions — erythema, pruritus, pain, irritation, bruising — in a substantial minority; arthralgia and myalgia; peripheral oedema; paraesthesia and other sensory symptoms; and changes in glucose measures. Discontinuation for adverse reactions was more frequent than with placebo. The injection-site reactions are reactions to the peptide and its formulation at the site; the joint, fluid and sensory effects are the known actions of growth hormone, released in greater amounts, and they are the same effects seen with somatropin at lower frequency and severity.

Reactions to the molecule

Local reactions at the site are the commonest and are attributed to the peptide, which is a foreign protein-like molecule, and to the formulation. Most were mild and resolved. Hypersensitivity reactions including rash and urticaria were reported in a small number and are a labelled warning. Antibody formation, described on its own page, did not correlate with local reactions.

These are the reactions a releasing-hormone analogue has that the hormone itself would not.

Reactions to the released hormone

Fluid retention producing oedema, arthralgia and carpal tunnel-like paraesthesia is the growth hormone signature, familiar from somatropin and from acromegaly. Glucose intolerance is the metabolic cost. All were less frequent than in earlier trials of growth hormone in the same condition, consistent with the bounded response of a releasing-hormone analogue, and they tended to appear early and diminish.

The label advises attention to glucose and to IGF-1.

Why it is here

Because the labelled adverse reactions are part of what is established about the molecule. It describes the label and no use of the pack.

Tesamorelin is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
