One gene on chromosome 20, a precursor of 108 residues, and expression well beyond the hypothalamus

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
The GHRH gene lies on the long arm of chromosome 20 and encodes a 108-residue preprohormone from which the signal peptide and flanking peptides are removed to give the 44-residue amidated hormone and, by alternative processing, the 40-residue carboxyl-terminally truncated form. Expression is highest in the arcuate nucleus of the hypothalamus, which supplies the pituitary. The gene is also expressed in placenta, where the peptide is found in large amounts, in the gastrointestinal tract and pancreas, in testis and ovary, and in lymphocytes, where a local growth-hormone-releasing or paracrine role has been proposed but is less established than the pituitary one.

The precursor

The preprohormone carries a signal peptide, an amino-terminal flanking region, the hormone itself and a carboxyl-terminal peptide. Prohormone convertases cut at paired basic residues to release the hormone, and a glycine donor residue is converted to the carboxyl-terminal amide. The 40-residue form arises from cleavage at an internal site and is fully active, since residues 30 to 44 are not required for receptor activation.

Tesamorelin corresponds to the full 44-residue product with one modification.

The extra sites

Placental GHRH is abundant and was once a candidate for the tumour-like production that gave the 1982 isolation; its role in pregnancy is unclear. Gut and pancreatic expression is low and of uncertain function. Lymphocyte expression, with lymphocyte expression of the receptor, underlies a literature on GHRH as an immune modulator that is largely in vitro. The hypothalamic-pituitary axis remains the one function that is fully established.

The receptor is likewise expressed outside the pituitary at low levels, including in some tumours, as the tumour page describes.

Why it is here

Because the gene and its expression pattern are part of what is established about the hormone the analogue copies. It describes genetics and no use of the pack.

Tesamorelin is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
