The other beta-thymosins, how they differ, and why one of them became a cancer marker

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
Three beta-thymosins are expressed in humans. Thymosin beta-4 is the most abundant and nearly ubiquitous. Thymosin beta-10 differs at about a quarter of its 43 positions, is prominent during development and in some tumours, and binds actin similarly. Thymosin beta-15, 45 residues, was identified in the 1990s as upregulated in metastatic prostate cancer cells and has been studied as a urinary marker. All three sequester actin through the same conserved motif and all are small, acidic, unstructured peptides. The family illustrates that the actin-binding function is ancient and conserved, and that expression of individual members tracks cell motility — which is why beta-15 appears in migrating cancer cells.

The differences

Sequence identity among the three is high and the actin-binding motif is conserved. Expression differs: beta-4 everywhere, beta-10 enriched in developing tissue and reduced in many adult tissues, beta-15 largely absent from normal adult tissue and induced in motile tumour cells. Their affinities for actin are similar within a few-fold.

Rodents express a beta-4 and a beta-10; the beta-15 gene is primate-specific in its present form.

Beta-15 and cancer

Bao and colleagues identified thymosin beta-15 in 1996 as a gene upregulated in rat prostate cancer lines selected for metastatic behaviour, and human studies since have associated its expression with prostate, breast and other cancers and tested it in urine as a marker. The association is with motility rather than with growth, consistent with the actin function.

The finding is also a reminder that factors promoting cell migration promote it in every cell type that migrates.

Why it is here

Because the family context explains what kind of protein thymosin beta-4 is and why migration-related effects cut both ways. It describes biology and no use of the pack.
