Follistatin: a binding protein, not a signalling peptide

Published 24 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
Follistatin is a secreted glycoprotein that binds members of the TGF-beta family — activins among them — and neutralises them by preventing receptor engagement. It is an antagonist that works by sequestration rather than by signalling through a receptor of its own.

Binding rather than signalling

Most compounds in a peptide catalogue act on a receptor. Follistatin does not: it binds its targets directly and holds them, so they cannot reach the receptors they would otherwise activate. The effect is subtraction rather than addition.

Nakamura and colleagues reported in 1990 that follistatin binds activin, which established the mechanism. Everything written about follistatin since rests on that: it is a sink for particular proteins.

What it binds

Activins, and other members of the transforming growth factor beta superfamily, with different affinities. The family is large and the binding is not uniform across it, which is why "follistatin blocks TGF-beta" is too loose a statement to be useful.

Which targets matter depends entirely on the system being studied, because what is present to be bound differs between tissues and between culture conditions.

Why that shapes an experiment

An antagonist that works by sequestration produces an effect proportional to how much of its target is around. In a system where the target is scarce, adding follistatin may do very little, and that is a property of the model rather than a failure of the material.

The composition of the system therefore bounds what the addition can produce in it. REVIVE LAB UAE offers no dosing or protocol guidance of any kind.
