What mass spectrometry confirms for IGF-1 LR3, and what it leaves to other methods

IGF-1 LR3 has an average molecular mass of about 9,111 daltons, against 7,649 for native IGF-1. In electrospray it appears as a charge envelope of perhaps five to ten states, narrower than a 22 kDa protein gives and wider than a peptide. Deconvolution gives the neutral mass, which confirms the full chain and that all three disulfides are formed; six daltons more would indicate a reduced protein. The swap isomer, with the same formula, is invisible to this test.

What the mass distinguishes

LR3 from native IGF-1, by about 1,460 daltons. The intact chain from truncations. Oxidised from unoxidised, by sixteen daltons per methionine, on a reasonably resolving instrument. Reduced from folded, by two daltons per disulfide. These are the common identity failures and intact mass covers them.

A 9 kDa protein is small enough that a high-resolution instrument resolves its isotopes, so monoisotopic mass can be used.

What it does not distinguish

Disulfide isomers. Deamidation, at one dalton, on a lower-resolution instrument. Conformational state. Aggregates, which dissociate in the source. Each of these is a separate method: peptide mapping, ion exchange, size exclusion.
