Three routes to the same kinase, and why the literature compares them

Published 26 October 2026 · REVIVE LAB UAE Research Desk · 5 min read
AMP-activated protein kinase is switched on when the ratio of AMP to ATP rises, or by molecules that mimic AMP. AICAR enters cells and is converted to ZMP, an AMP analogue that binds the kinase directly. Metformin inhibits complex I of the respiratory chain, lowers ATP and raises AMP, activating the kinase indirectly. MOTS-c, by the 2015 proposal, blocks the folate cycle so that endogenous AICAR accumulates — a route that converges on the AICAR mechanism from inside the cell. The three are compared because they reach one target by different doors.

AICAR

A purine precursor, available as a reagent since the 1990s and used as the standard pharmacological AMPK activator in cell and animal studies. It is also a doping concern, listed as a metabolic modulator. Its weakness as a tool is that ZMP has effects beyond AMPK.

The MOTS-c papers used it as a positive control.

Metformin

The most widely used oral medicine for type 2 diabetes, whose AMPK activation was described in 2001 and is one of several proposed mechanisms. It acts on mitochondria to shift the energy state rather than on the kinase itself. Its effects on glucose output from the liver are its clinical basis.

It appears in MOTS-c papers as the clinical reference point for an AMPK-linked metabolic agent.

MOTS-c

Proposed to raise endogenous AICAR by blocking the folate cycle, so that the cell makes its own AMP mimic. If correct, it is a third door: not an exogenous analogue and not an energy-state change, but a metabolic diversion. The target that is blocked has not been identified. The comparison is instructive and the mechanism remains a proposal.

MOTS-c is supplied by REVIVE LAB UAE for laboratory research use only, with identity by mass spectrometry and purity by HPLC for the batch. It is not for human or veterinary use, and no dosing, administration or protocol guidance is provided.
